In Vitro and In Silico Pharmacological Screening and Evaluation: Advantages and Disadvantages

Pharmacological screening and evaluation are essential steps in the drug discovery and development process. They help researchers identify promising drug candidates, determine their biological activity, understand their mechanism of action, and assess their potential safety before further development. Among modern screening approaches, in vitro and in silico methods are widely used because they can provide valuable information at an early stage and help reduce the time, cost and experimental burden of drug discovery.

In vitro pharmacological screening involves testing compounds outside a living organism using isolated enzymes, receptors, cells, tissues or other biological systems. It allows researchers to study specific pharmacological effects under controlled conditions and provides useful information about drug–target interactions, cellular responses and mechanisms of action. However, in vitro models cannot completely reproduce the complex physiological environment of a living organism, and therefore their results may not always translate directly to in vivo effects.

In silico pharmacological screening uses computational techniques such as molecular docking, virtual screening, QSAR and ADMET prediction to predict the activity and properties of drug candidates. It enables rapid screening of large numbers of compounds and helps researchers prioritise the most promising molecules for experimental testing. Nevertheless, computational predictions depend on the quality of the data, models and assumptions used and must be experimentally validated.

Understanding the advantages and limitations of in vitro and in silico approaches is therefore important for pharmacy students and competitive-examination aspirants.
